چکیده

Tetrahymena geleii (S) (Colpidium camp&m) is a ciliated protozoan which has been shown to be capable of synthesizing lipides from protein at an unusually high rate (1, 2). The next obvious step in studies concerned with the metabolism of this organism, therefore, seemed to be a search for the compound, or compounds, serving as precursor of the lipide material. Acetate is known to play a key r&e in the metabolism of lipides, carbohydrates, and proteins (7). Rittenberg and Bloch (8, 9) and Ponticorvo, Rittenberg, and Bloch (10) have shown that acetate may condense to form fatty acids in mammalian tissue. Smedley-MacLean and Hoffert (11) and White and Werkman (12) have demonstrated that in yeast, also, the intact acetate molecule is converted to lipide material. It has also been demonstrated (13-16) that acetate may condense to form higher, non-reducing carbohydrates. The classical work of Schoenheimer and Rittenberg (17) demonstrated that carbohydrate may be transformed into lipides. It therefore seemed of interest to investigate the fate of acetate utilization by T. geleii (S) in an attempt to ascertain whether acetate can serve as a precursor of lipides, and, if so, whether it must first be converted to carbohydrate.

Utilization of guanine for nucleic acid biosynthesis by Tetrahymena geleii.

As a result of the investigations of Kidder and coworkers, it has recently become possible to grow the ciliated prot.oaoan Tetrahymena geleii in a culture medium which is of chemically known composition (1, 2), as well as free of other microorganisms. Interest was aroused by similarities in its requirements for amino acids (2, 3) and vitamins (4) to those of the mammal, although differences hav...

The Effects of Oxygen, Carbon Dioxide, and Pressure on Growth in Chilomonas Paramecium and Tetrahymena Geleii Furgason

1. The effects of O(2), CO(2), and pressure were studied in two very different species of protozoa, a flagellate, Chilomonas paramecium, grown in acetate-ammonium solution and a ciliate, Tetrahymena geleii, grown in 2 per cent proteose-peptone solution. 2. Chilomonas and Tetrahymena live and reproduce in solutions exposed to a wide range of O(2) concentrations, but Chilomonas is killed at high ...

Growth studies on Tetrahymena geleii H.

l’ctmhymena g&ii II, a holotrichous protozoan, was isolated as a bacteria-free culture in 1931 by Dr. Alford Hetherington at Pacific Grove, California. The culture of the ciliate employed in the present studies was obtained in 1945 through the courtesy of Dr. George W. Kidder, who had received a culture from Dr. R. H. Hall in 1938 (2). Hetherington named this microorganism Colpidium campylum, b...

Enzyme Systems in Tetrahymena Geleii S

The rates of activity of the dehydrogenase systems in Tetrahymena, which are concerned with carbohydrate oxidation, in descending order of activity are: lactic > isocitric > succinic = glucose > glucose-6-phosphate = 6-phosphogluconic = malic > glutamic = cytochrome linked alpha-glycerophosphate dehydrogenase. No evidence was obtained to indicate the presence of DPN linked alpha-glycerophosphat...

The amino acid composition and nitrogen metabolism of Tetrahymena geleii.

The nitrogen composition and metabolism of the typical animal microorganisms are relatively unknown. This is true largely because of the past unavailability of the Protozoa either in pure culture or in synthetic media. The ciliated protozoan, Tetrahymena geleii, is unique in that it has been available in pure culture for some time and recently (1949) it has been grown in synthetic medium (2). I...
